Bacillus Megaterium Extracción Purificación Caracterización
Environmental pollution caused by the accumulation of petroleum-based plastics has aimed at finding of biodegradable materials such as PHAs. Polyhydroxybutyrate (PHB) is produced by various microorganisms as Bacillus megaterium from cheap carbon sources such as crude glycerol. One of the key aspects of the production process is the extraction and purification of the polymer. This paper presents a methodology for extracting PHB produced by Bacillus megaterium B2 evolved from crude glycerol as sole carbon source using doudecilsulfato sodium (SDS) and digesting agent
evaluating different pretreatment options and the contribution of each. Tests conducted included: A pretreatment (without pretreatment

cell lysis with NaOH and NaClO) followed
SDS digestion
heat treatment
washing with distilled water
solvent washing and drying. PHB extracted was characterized by thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). The higher purity of the polymer is obtained using NaClO as pretreatment
it is additionally noted that the stage with the greatest impact in the extraction was washed with distilled water after heat treatment. The melting temperatures (Tm) and degradation (Td) obtained for PHB are extracted within the range reported in the literature for this polymer.
