Triosephosphatase isomerase (tpi) housekeeping gene - coupled with RFLP approach was done for isolates identification, and further reference primers were used for toxinotype evaluation of 17 isolates expressing proteins in supernatant. C. perfringens isolates expressing alpha toxin were found in soil samples from Nemocón, Cundinamarca and isolate from Barrancas (Guajira) amplified for reference alpha toxin of C. perfringens and Clostridium septicum. Toxins sequence variability was demonstrated in comparison to public database strains available in NCBI; the design of primers for further molecular characterization of local isolates is suggested, along with phylogenetic analysis based on MLST housekeeping genes.

Los protocolos del Manual de Análisis Bacteriológico (BAM) de la FDA se utilizaron para el aislamiento de los dos microorganismos clostridiales más representativos en Colombia: C. botulinum y Clostridium perfringens. El gen tpi (triosfosfatasa isomerasa), junto con la técnica RFLP (Restriction Fragment Length Polymorphism) se utilizó para la identificación de los aislamientos. Se emplearon diferentes primers de referencia para la identificación por toxinotipo de 17 aislamientos que demostraron expresar proteínas asociadas a toxina. De los 67 suelos analizados, se encontraron aislamientos de C. perfringens que expresan alfa toxina provenientes de Nemocón, Cundinamarca y aislamientos de Barrancas (Guajira) que amplifican para la alfa toxina de referencia de C. perfringens y Clostridium septicum. La variabilidad en las secuencias nucleotídicas de toxinas fue demostrada en comparación con cepas de bases de datos públicas disponibles en NCBI; se sugiere el diseño de primers locales que permitan caracterizar molecularmente los aislamientos locales.
