description |
Several outbreaks since 1973 have been observed in Colombian cattle with a presumptive diagnosis of Clostridium botulinum diseases [1]. FDA Bacteriological Analytical Manual protocols were used for the isolation of the two representative clostridia microorganisms in Colombia: C. botulinum and C. perfringens. Triosephosphatase isomerase (tpi) housekeeping gene - coupled with RFLP approach was done for isolates identification, and further reference primers were used for toxinotype evaluation of 17 isolates expressing proteins in supernatant. C. perfringens isolates expressing alpha toxin were found in soil samples from Nemocón, Cundinamarca and isolate from Barrancas (Guajira) amplified for reference alpha toxin of C. perfringens and Clostridium septicum. Toxins sequence variability was demonstrated in comparison to public database strains available in NCBI; the design of primers for further molecular characterization of local isolates is suggested, along with phylogenetic analysis based on MLST housekeeping genes. |
