The first part of the current document (Theoretical framework and Objectives) defines the main concepts involved in the biotechnological potentials of bacteria, focusing on the case of L. sphaericus and its promising potential applications for biocontrol and bioremediation. In addition, the controversy over the species circumscription of L. sphaericus is reviewed. Lastly, this section describes the state of the art and establishes the objectives of the current study. In Chapter 1, the parameters and results of the genome sequencing of the entomopathogenic strains 2362 and OT4b.49 are described. Results from these genome sequencing are compared to the current available L. sphaericus genomes on the NCBI Genome database. In Chapter 2, the species circumscription of L. sphaericus is discussed. A new bacterial species made up of the toxic strains is suggested. This propose is supported by the genomic data obtained in this study and by the experimental evidence previously reported in the scientific literature. In Chapter 3, a metabolic reconstruction of the L. sphaericus toxic strains is presented. This reconstruction was built from the obtained genomic data and offers a guideline for further studies on the biotechnological implementation of L. sphaericus. Finally, the last part of this document shows the Concluding remarks of the current work, including the questions about the L. sphaericus biology that remains unanswered. The Appendices section contains some unpublished results and other supplemental material that bolsters the hypotheses proposed herein. |

Tal proyecto tuvo como objetivo alcanzar un entendimiento de la biología de L. sphaericus como especie y agente en biotecnología desde un perspectiva genómica. La primera parte de este documento define los conceptos claves relacionados con los potenciales biotecnológicos de las bacterias, enfocándose en el caso de L. sphaericus y sus aplicaciones en control biológico y remediación ambiental. Adicionalmente, se describe la controversia taxonómica que actualmente engloba a esta especie y se establecen los objetivos del presente estudio. En el capítulo 1, se detallan los parámetros y resultados de la secuenciación de los genomas de las cepas entomotóxicas 2362 y OT4b.49 en relación a los genomas actualmente disponibles en las bases de datos del NCBI para esta especie. En el capítulo 2, se discute la circunscripción de especie de L. sphaericus. Se sugiere que las cepas tóxicas conforman una especie separada del resto de las cepas, que no poseen una circunscripción clara. Esta hipótesis se soporta con los datos genómicos obtenidos en este estudio así como en evidencia experimental reportada en la literatura. En el capítulo 3, se presenta una reconstrucción metabólica del linaje tóxico de L. sphaericus, donde se resaltan sus potenciales biotecnológicos y se proponen lineamientos para su implementación. Finalmente, en la última parte del documento, se plantean las conclusiones del presente trabajo así como se proponen las preguntas sobre la biología de esta especie que permanecen aún sin respuesta.
