description La invasion de esporozoitos de Plasmodium spp a hepatocitos humanos representa el primer paso para que se establezca la invasion en el humano, esto ocurre mediante la interacción de proteínas de la superficie del esporozoito y receptores en la superficie del hepatocito. La proteína anónima relacionada con trombospondina de Plasmodium vivax (PvTRAP) es un polipéptido presente en la superficie de esporozoítos, del cual en otras especies de Plasmodium se ha investigado su participación en el proceso de invasión al hepatocito. Esta proteína es conservada en varias especies del género Plasmodium caracterizándose por la presencia de un dominio de repetición de trombospondina tipo I (TSR) y un dominio A similar al Factor von Willebrand (vW). En el presente estudio se amplificaron dos fragmentos del gen pvtrap que codifican para el ectodominio de la proteína PvTRAP y los dominios TSR-vW. Cada producto fue clonado en el vector pFastBac-HT-C y secuenciado mediante la técnica de Sanger, obteniendo por primera vez la secuencia de nucleótidos y aminoácidos de la proteína PvTRAP derivada de la cepa VCG-1. Los clonos recombinantes confirmados fueron utilizados para la obtención de baculovirus recombinantes en células de insecto Sf9. Una alta concentración de stock viral fue utilizada para la expresión de los dos fragmentos recombinantes: PvTRAP y PvTRAP-vW-TSR. El ectodominio PvTRAP se purificó mediante cromatografía de afinidad, se dializó exhaustivamente frente a PBS y se cuantificó. Mediante ensayos de citometría de flujo se encontró que la proteína PvTRAP se unió a la línea celular hepática ATCC HB-8065. Futuros estudios son necesarios para evaluar las regiones mínimas de interacción con el fragmento PvTRAP-vW-TSR. Los resultados de este estudio destacan la capacidad de PvTRAP para interactuar con la célula blanco de P. vivax y pone en evidencia el potencial de esta polipéptido en P. vivax como ha sido mostrado para P. falciparum.

La proteína anónima relacionada con trombospondina de Plasmodium vivax (PvTRAP) es un polipéptido presente en la superficie de esporozoítos, del cual en otras especies de Plasmodium se ha investigado su participación en el proceso de invasión al hepatocito. Esta proteína es conservada en varias especies del género Plasmodium caracterizándose por la presencia de un dominio de repetición de trombospondina tipo I (TSR) y un dominio A similar al Factor von Willebrand (vW). En el presente estudio se amplificaron dos fragmentos del gen pvtrap que codifican para el ectodominio de la proteína PvTRAP y los dominios TSR-vW. Cada producto fue clonado en el vector pFastBac-HT-C y secuenciado mediante la técnica de Sanger, obteniendo por primera vez la secuencia de nucleótidos y aminoácidos de la proteína PvTRAP derivada de la cepa VCG-1. Los clonos recombinantes confirmados fueron utilizados para la obtención de baculovirus recombinantes en células de insecto Sf9. Una alta concentración de stock viral fue utilizada para la expresión de los dos fragmentos recombinantes: PvTRAP y PvTRAP-vW-TSR. El ectodominio PvTRAP se purificó mediante cromatografía de afinidad, se dializó exhaustivamente frente a PBS y se cuantificó. Mediante ensayos de citometría de flujo se encontró que la proteína PvTRAP se unió a la línea celular hepática ATCC HB-8065. Futuros estudios son necesarios para evaluar las regiones mínimas de interacción con el fragmento PvTRAP-vW-TSR. Los resultados de este estudio destacan la capacidad de PvTRAP para interactuar con la célula blanco de P. vivax y pone en evidencia el potencial de esta polipéptido en P. vivax como ha sido mostrado para P. falciparum.Resumen 9 Introducción 10 1.
