description La mieloperoxidasa (MPO) es una proteína antimicrobiana presente en los gránulos azurófilos de los neutrófilos, su acción recae cuando se conjuga en el fagosoma con peróxido de hidrógeno, resultando en sustancias tóxicas para los microorganismos fagocitados (1). Se ha descrito que las infecciones vulvovaginales recurrentes por Candida spp tienen relación con la deficiencia priMaría de MPO, el cual es un trastorno autosómico recesivo (2), que presenta polimorfismos asociados al procesamiento postraduccional defectuoso de la proteína precursora o con errores pretraduccionales (3), por esto, el propósito fue identificar la relación que existe entre dos polimorfismos de la MPO (Y173C y M251T) con una mayor probabilidad de presentar este tipo de infecciones, puesto que este tipo de polimorfismos no han sido estudiados en el país. Frente a la metodología, se realizó una encuesta, donde se recolectaron 27 muestras de mujeres entre 20-45 años, se les realizó cuadro hemático, tinción Wright y tinción de peroxidasa leucocitaria. Se prosiguió con la extracción de ADN utilizando el kit Wizard® Genomic DNA Purification de Promega, luego se realizó la cuantificación del ADN extraído por espectrofotometría en NanoDrop, y la evaluación de la integridad del ADN mediante electroforesis. Por otro lado, se diseñaron los primers haciendo uso de herramientas bioinformáticas para la estandarización de la PCR convencional, posteriormente se usaron enzimas de restricción y se secuenciaron los productos amplificados con el fin de identificar los polimorfismos mencionados previamente.

Se ha descrito que las infecciones vulvovaginales recurrentes por Candida spp tienen relación con la deficiencia priMaría de MPO, el cual es un trastorno autosómico recesivo (2), que presenta polimorfismos asociados al procesamiento postraduccional defectuoso de la proteína precursora o con errores pretraduccionales (3), por esto, el propósito fue identificar la relación que existe entre dos polimorfismos de la MPO (Y173C y M251T) con una mayor probabilidad de presentar este tipo de infecciones, puesto que este tipo de polimorfismos no han sido estudiados en el país. Frente a la metodología, se realizó una encuesta, donde se recolectaron 27 muestras de mujeres entre 20-45 años, se les realizó cuadro hemático, tinción Wright y tinción de peroxidasa leucocitaria. Se prosiguió con la extracción de ADN utilizando el kit Wizard® Genomic DNA Purification de Promega, luego se realizó la cuantificación del ADN extraído por espectrofotometría en NanoDrop, y la evaluación de la integridad del ADN mediante electroforesis. Por otro lado, se diseñaron los primers haciendo uso de herramientas bioinformáticas para la estandarización de la PCR convencional, posteriormente se usaron enzimas de restricción y se secuenciaron los productos amplificados con el fin de identificar los polimorfismos mencionados previamente.TABLA DE CONTENIDO RESUMEN. 12 INTRODUCCIÓN . 13 1.
