Brucellosis and bovine tuberculosis (TBB) are infectious diseases that significantly impact Colombian livestock production. The previous situation persists because control strategies are ineffective with failures in the early detection of cases and adequate surveillance of these diseases. The objective of this work is to present a comprehensive strategy for the early detection and mapping of brucellosis and TBB cases. A protocol for bacterial culture, genomic DNA extraction and amplification of specific Brucella abortus and Mycobacterium bovis genes was standardized for their specific detection in raw milk and artisan cheese samples. 5 raw milk samples and 5 cheese samples were evaluated. Genomic DNA was obtained at an average concentration of 64.3 ng/µL and appropriate purity (average A260/A280 ratio of 1.6). In 4 cheese samples the 16S ribosomal genes were amplified by PCR and 1 cheese sample was positive for Mycobacterium bovis. An interactive web tool was designed with the cases of brucellosis and TBB between 2004-2020 reported to the Instituto Colombiano Agropecuario that presents the geographical distribution of foci on the time scale at the national level. The identification of M. bovis in dairy products suggests that the pathogen could be present in dairy products that are sold locally. |

The identification of M. bovis in dairy products suggests that the pathogen could be present in dairy products that are sold locally.La brucelosis y la tuberculosis bovina (TBB) son enfermedades infecciosas que impactan significativamente la producción ganadera colombiana. La anterior situación persiste porque las estrategias de control son ineficaces con fallas en la detección temprana de casos y adecuada vigilancia de dichas enfermedades. El objetivo de este trabajo es presentar una estrategia integral para la detección temprana y mapeo de casos de brucelosis y TBB. Se estandarizó un protocolo de cultivo bacteriano, extracción de ADN genómico y amplificación de genes específicos de Brucella abortus y Mycobacterium bovis para su detección específica en muestras de leche cruda y queso artesanal. Se evaluaron 5 muestras de leche cruda y 5 muestras de queso. Se obtuvo ADN genómico a una concentración promedio de 64,3 ng/µL y pureza apropiada (relación A260/A280 promedio de 1,6). En 4 muestras de queso se amplificaron por PCR los genes ribosomales 16S y 1 muestra de queso fue positiva para Mycobacterium bovis. Se diseñó una herramienta web interactiva con los casos de brucelosis y TBB entre 2004-2020 reportados al Instituto Colombiano Agropecuario que presenta la distribución geográfica de focos en la escala de tiempo a nivel nacional.
