From a collection of Bacillus thuringiensis strains isolated in Colombia from plants and exhibiting toxic activity against lepidopteran insects, 310 bacterial isolates were tested to determine lactonase activity by using biosensor systems in presence of synthetic N-hexanoyl-L-homoserine lactone (C6-HSL) and N-octanoyl-L-homoserine lactone (C8-HSL). From them, 251 strains showed degrading activity to both C6-HSL and C8-HSL, 57% exhibited degrading activity to C6-HSL and 43% to C8-HSL. One B. thuringiensis strain, denoted as 147-115-16, that exhibit high degrading activity to C6-HSL and C8-HSL, was able to attenuate soft rot symptoms in infected potato slices with Pectobacterium carotovorum. This strain contains an homologous of the aiiA gene that was cloned, sequenced and expressed in Esherichia coli DE3. The recombinant protein AiiA147-11516 display activity to C6-HSL, C8-HSL, N-(β-ketocaproyl) (3-O-C6-HSL) and N-3-oxo-dodecanoyl (3-O-C12-HSL). The recombinant strain in the presence of P. caratovorum cultures was able to attenuate the infection, suggesting that it interferes either on the accumulation or response to the AHLs signals. Acording to this data and based on previous report from recombinant AiiA147-11516, this enzyme exhibit activity to wide range of catalytic substrates suggesting its industrial application in the disease control programs by plants transformation. XVI No. 1 Julio, 2014. 153-162Derechos Reservados - Revista Colombiana de Biotecnología, 2014info:eu-repo/semantics/openAccessAtribución-NoComercial 4.0 Internacional (CC BY-NC 4.0)https://creativecommons.org/licenses/by-nc/4.0/http://purl.org/coar/access_right/c_abf2http://www.scielo.org.co/pdf/biote/v16n1/v16n1a19.pdfLactonesQuorum sensingQuorum quenchingLactonasesPectobacterium caratovorumIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strainIdentificación, clonación y actividad lactonasa de la proteína recombinante de N-ácil homoserina lactonasa (AiiA) de Bacillus thuringiensis cepa 147-115-16Artículo de revistahttp://purl.org/coar/resource_type/c_6501http://purl.org/coar/resource_type/c_2df8fbb1Textinfo:eu-repo/semantics/articlehttp://purl.org/redcol/resource_type/ARTinfo:eu-repo/semantics/publishedVersionhttp://purl.org/coar/version/c_970fb48d4fbd8a85PublicationORIGINALIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strain.pdfIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strain.pdfapplication/pdf137948https://repositorio.udes.edu.co/bitstreams/7a857374-4571-4983-a84a-9426c6e72a6b/downloadfd9f5388ec0327d3f8cf53ca5daf70dbMD55LICENSElicense.txtlicense.txttext/plain; charset=utf-859https://repositorio.udes.edu.co/bitstreams/c6e01906-0b70-4eef-9443-b36e53d68c15/download38d94cf55aa1bf2dac1a736ac45c881cMD52TEXTIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16.pdf.txtIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16.pdf.txtExtracted texttext/plain44402https://repositorio.udes.edu.co/bitstreams/123f7e69-4b60-47b8-a114-bbf2b486ad31/download58f1fcb8aed1da3b46fbeb26fb77233cMD53Identification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strain.pdf.txtIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strain.pdf.txtExtracted texttext/plain2https://repositorio.udes.edu.co/bitstreams/156c1149-a1a8-46ec-8032-12afb3d60217/downloadd784fa8b6d98d27699781bd9a7cf19f0MD56THUMBNAILIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16.pdf.jpgIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16.pdf.jpgGenerated Thumbnailimage/jpeg1545https://repositorio.udes.edu.co/bitstreams/86402029-1776-45fe-9e8c-1ebb3b850fca/download4acde44de7ff0e7e27ea37d5a8ff289eMD54Identification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strain.pdf.jpgIdentification, cloning and lactonase activity of recombinant protein of N-acyl homoserine lactonase (AiiA) from Bacillus thuringiensis 147-115-16 strain.pdf.jpgGenerated Thumbnailimage/jpeg1339https://repositorio.udes.edu.co/bitstreams/ca6aaf9b-e96b-42e7-9aab-22941eb66bd7/downloadddb57e08c9c81f9d25c4a0a922e0217cMD57001/3579oai:repositorio.udes.edu.co:001/35792023-10-10 08:34:29.432https://creativecommons.org/licenses/by-nc/4.0/Derechos Reservados - Revista Colombiana de Biotecnología, 2014https://repositorio.udes.edu.coRepositorio Universidad de Santandersoporte@metabiblioteca.comTGljZW5jaWEgZGUgUHVibGljYWNpw7NuIFVERVMKRGlyZWN0cmljZXMgZGUgVVNPIHkgQUNDRVNPCgo= |
