Se emplearon 13 perros adultos, de ambos sexos, clínicamente sanos con edades entre 2 y 7 años y un peso igual o superior a 20kg. Estos ingresaron al área de clínica quirúrgica del Hospital Veterinario de la Universidad de Caldas para un procedimiento electivo. A cada canino se le tomó una muestra de 34 ml de sangre entera divida en seis tubos; uno con anticoagulante EDTA que fue empleado para realizar conteo celular basal y cinco tubos sin anticoagulante, pero con sílice micronizado que fueron a su vez divididas en tres grupos los cuales recibieron diferentes tipos de procesos y fueron llamados protocolo de centrifugación Nº1 (2 tubos) a 410g durante 10 minutos, descrito por Dohan et al. para obtener fibrina rica en plaquetas en la especie humana, protocolo de centrifugación Nº2 (2 tubos) a 191g durante 6 minutos, protocolo descrito por Silva et al. para obtener concentrado autólogo de plaquetas en la especie canina; y el denominado control (1 tubo) que fue dejado en reposo para obtener una comparación posterior entre estos mediante microscopia electrónica de barrido y un conteo automatizado del sobrenadante residual de cada tubo. Evaluar la distribución celular dentro del coágulo no fue posible debido a la característica gelatinosa que presenta la estructura y que envuelve las células, en este caso solo se pudo comprobar su presencia por visualización directa de cuerpos celulares parcialmente atrapados o adheridos a la superficie del coagulo. Sin embargo, al realizar conteo hematológico al sobrenadante de cada tubo se pudo determinar que existe una diferencia estadísticamente significativa del coagulo generado en reposo también denominado control, respecto a los coágulos generados por los protocolos 1 y 2 en términos de eritrocitos. El análisis de las demás estructuras celulares demostró que no existe una diferencia estadísticamente significativa entre estos. Por otro lado La manipulación de la sangre mediante los dos tipos de protocolos si permitió la obtención de un coagulo y una malla estable y resistente, con características visco elásticas deseables al momento de una aplicabilidad clínica. Sin embargo, el tamaño del coagulo generado con el protocolo Nº1 logra una estructura de 1cm a 1.5cm más largo, siendo este tipo de estructura más deseable cuando se enfrentan lesiones de gran tamaño o con grandes pérdidas de tejido.eng: The purpose of this study was to standardize and describe a method for the formation of a platelet-rich fibrin clot or mesh in canines that was molecularly stable for characterization and possible clinical application. Thirteen adult dogs, of both sexes, we`re used clinically healthy with ages between 2 and 7 years and a weight equal to or greater than 20 kg. These entered the surgical clinic area of the Veterinary Hospital of the University of Caldas for an elective procedure. Each canine is taken a 34 ml sample of whole blood divided into six tubes; one with EDTA anticoagulant that was used to perform a basal cell count and five tubes without anticoagulant, but with micronized silica that were in turn divided into three groups which had different types of processes and were called centrifugation protocols Nº1 (2 tubes) at 410 g for 10 minutes, described by Dohan et al. to obtain platelet-rich fibrin in the human species, centrifugation protocol Nº2 (2 tubes) at 191g for 6 minutes, protocol labeled by Silva et al. to obtain autologous platelet concentrate in the canine species; and the so-called control (1 tube) that was left to rest to obtain a subsequent comparison between these using scanning electron microscopy and an automated count of the residual supernatant of each tube. Evaluating the cellular distribution within the clot was not possible due to the gelatinous characteristic that the structure presents and that surrounds the cells. In this case, its presence could only be detected by direct visualization of the cell bodies trapped or adhering to the surface of the clot. However, by performing a hematological count on the supernatant of each tube, it was possible to determine that there is a statistically significant difference between the resting clot, also called control, with respect to the clots generated by protocols 1 and 2 in terms of erythrocytes. The analysis of the other mobile structures showed that there is no statistically significant difference between them. On the other hand, the manipulation of the blood by means of the two types of protocols if obtaining a clot and a stable and resistant mesh, with desirable elastic visual characteristics at the time of clinical applicability.
