Blastocystis es un microeucariota intestinal que ha suscitado atención debido a su amplia distribución en animales y seres humanos. El riesgo de circulación zoonótica surge principalmente del contacto estrecho con animales infectados. Por lo tanto, el siguiente estudio tuvo como objetivo evaluar la diversidad y frecuencia de los subtipos de Blastocystis en muestras humanas y animales colombianas utilizando la secuenciación completa del gen 18S rRNA. Para este propósito, 341 muestras fecales humanas y 277 muestras fecales animales (de bovinos, ovinos, caprinos, cerdos, gatos y perros), fueron recolectadas de diferentes regiones colombianas y analizadas usando detección basada en PCR y secuenciación de próxima generación (NGS) del gen 18S SSU rRNA de longitud completa. Entre las 618 muestras de ambos huéspedes, humanos y animales, los resultados revelaron una frecuencia generalizada de Blastocystis, con un 48,09% (n=164) en humanos y un 31,4% (n=87) de detección en animales. Perros, gatos, ovejas, cerdos y animales salvajes dieron positivo, en consonancia con los patrones de prevalencia mundial. Además, se secuenciaron 29 muestras humanas y 23 muestras animales mediante tecnología ONT, a partir de las cuales se generaron 11 secuencias únicas de lectura larga que se agruparon con sus secuencias de referencia comparadas. La distribución de subtipos varió dentro de los hospedadores, detectándose ST1 y ST3 tanto en muestras humanas como animales. Los subtipos ST5, ST10, ST14, ST15, ST21, ST24, ST25 y ST26 se limitaron a hospedadores animales, algunos de los cuales se considera que tienen potencial zoonótico. Por otra parte, el ST2 se encontró exclusivamente en muestras humanas de la región de Bolívar. Se produjeron infecciones mixtas tanto en animales como en humanos, 60,86% y 27,58% respectivamente. Además, hasta donde sabemos, este es el primer estudio en Colombia que identifica ST15 en cerdos y ST25 en ovejas. Los subtipos (STs) identificados en este estudio indican que ciertos animales pueden servir como reservorios con potencial de transmisión zoonótica. La identificación de subtipos zoonóticos pone de relieve el uso de la secuenciación de nueva generación, ya que la profundidad y la resolución de las secuencias aumentan, lo que permite comprender mejor los ST de importancia médica y veterinaria. También revela la coexistencia de diversos subtipos entre hospedadores. Es esencial seguir investigando para comprender la dinámica de transmisión, las implicaciones sanitarias y las estrategias de detección de Blastocystis en animales y humanos, sobre todo por el papel de los animales como reservorios y su estrecha interacción con los humanos. |

El riesgo de circulación zoonótica surge principalmente del contacto estrecho con animales infectados. Por lo tanto, el siguiente estudio tuvo como objetivo evaluar la diversidad y frecuencia de los subtipos de Blastocystis en muestras humanas y animales colombianas utilizando la secuenciación completa del gen 18S rRNA. Para este propósito, 341 muestras fecales humanas y 277 muestras fecales animales (de bovinos, ovinos, caprinos, cerdos, gatos y perros), fueron recolectadas de diferentes regiones colombianas y analizadas usando detección basada en PCR y secuenciación de próxima generación (NGS) del gen 18S SSU rRNA de longitud completa. Entre las 618 muestras de ambos huéspedes, humanos y animales, los resultados revelaron una frecuencia generalizada de Blastocystis, con un 48,09% (n=164) en humanos y un 31,4% (n=87) de detección en animales. Perros, gatos, ovejas, cerdos y animales salvajes dieron positivo, en consonancia con los patrones de prevalencia mundial. Además, se secuenciaron 29 muestras humanas y 23 muestras animales mediante tecnología ONT, a partir de las cuales se generaron 11 secuencias únicas de lectura larga que se agruparon con sus secuencias de referencia comparadas. La distribución de subtipos varió dentro de los hospedadores, detectándose ST1 y ST3 tanto en muestras humanas como animales. Los subtipos ST5, ST10, ST14, ST15, ST21, ST24, ST25 y ST26 se limitaron a hospedadores animales, algunos de los cuales se considera que tienen potencial zoonótico. Por otra parte, el ST2 se encontró exclusivamente en muestras humanas de la región de Bolívar. Se produjeron infecciones mixtas tanto en animales como en humanos, 60,86% y 27,58% respectivamente. Además, hasta donde sabemos, este es el primer estudio en Colombia que identifica ST15 en cerdos y ST25 en ovejas. Los subtipos (STs) identificados en este estudio indican que ciertos animales pueden servir como reservorios con potencial de transmisión zoonótica. La identificación de subtipos zoonóticos pone de relieve el uso de la secuenciación de nueva generación, ya que la profundidad y la resolución de las secuencias aumentan, lo que permite comprender mejor los ST de importancia médica y veterinaria. Therefore, the following study aimed to evaluate the diversity and frequency of Blastocystis subtypes in Colombian human and animal samples using complete sequencing of the 18S rRNA gene. For this purpose, 341 human stool samples and 277 animal fecal samples (from cattle, sheep, goat, pigs, cats, and dogs), were collected from different Colombian regions and analyzed using PCR-based detection and full-length 18S SSU rRNA gene Next-Generation Sequencing (NGS). Among the 618 samples from both hosts, humans and animals, the results revealed widespread Blastocystis frequency, with 48.09% (n = 164) in humans and 31.4% (n = 87) detection in animals. Dogs, cats, sheep, pigs, and wild animals tested positive, aligning with global prevalence patterns. Also, 29 human samples and 23 animal samples were sequenced using ONT technology from which 11 long-read unique sequences were generated and cluster with their compared reference sequences. The subtype distribution varied within hosts, detecting ST1 and ST3 in both human and animal samples. Subtypes ST5, ST10, ST14, ST15, ST21, ST24, ST25 and ST26 were limited to animals hosts, some of which are considered to have zoonotic potential. On the other hand, ST2 was found exclusively in human samples from Bolivar region. Mixed infections occurred in both animal and humans, 60.86% and 27.58% respectively. Moreover, to our knowledge, this is the first study in Colombia identifying ST15 in pigs and ST25 in sheep. The subtypes (STs) identified in this study indicate that certain animals may serve as reservoirs with the potential for zoonotic transmission. The identification of zoonotic subtypes highlights the use of Next Generation Sequencing as the depth and resolution of the sequences increases providing insights into STs of medical and veterinarian significance.
