description The Prostate Specific Antigen (PSA) generally is used quantitatively to detect and to watch over the development of prostate of cancer by serum levels of this protein, but also is found in high concentrations in semen. Different sensitive, reproducible and simple methods have been developed for the analysis of the presence of PSA including inmunocromatografy techniques. The procedures most common in the forensic detection of semen have focused in the detection of spermatozoa, inmunoelectroforetic activity of acid phosphatase and methods for the PSA detection. Although these methods has been used by many years, but some problems are associated with each method. The objective of this investigation was to validate the inmunocromatografy test in the detection of PSA in forensic evidences. Different dilutions of semen standar (100.000 ng/ml of PSA) were made, determining the low limit detection in 10 ng/ml. Positive results were obtained since concentrations of 500ng/ml in stains and swabs. The performance of the test was evaluated in the presence of interferences, such as blood, male and female urine, maternal milk, vaginal secretions, feces and saliva. It shows positive results in samples of blood and male urine with prostatic cancer. We reported a sensitivity of 91.6 % and a specificity of 100 %. These results suggest that this presumtive test can be usefull as preliminar test to determinate presence of semen in forensic samples.

These results suggest that this presumtive test can be usefull as preliminar test to determinate presence of semen in forensic samples.El antígeno prostático específico (PSA) es generalmente usado para detectar y monitorear cuantitativamente el desarrollo de cáncer de próstata por niveles en suero de esta proteína, esta también es encontrada en altas concentraciones en semen. Diversos métodos reproducibles, sensibles y simples han sido desarrollados para el análisis de la presencia de PSA incluyendo técnicas inmunocromatográficas. Los procedimientos más comunes para la detección forense de semen se han enfocado en la identificación de espermatozoides, actividad de la fosfatasa ácida y métodos inmunoelectroforéticos. Aunque estos métodos han sido utilizados por muchos años, existen algunos problemas asociados con cada método. El objetivo de esta investigación fue validar la prueba inmunocromatográfica para la detección de PSA en muestras forenses, para ello se realizaron diferentes diluciones de un patrón de semen con una concentración de PSA de 100.000ng/ml, encontrándose un límite de detección de 10 ng/ml. En manchas secas y escobillones, se presentaron resultados positivos a partir de concentraciones de 500 ng/ml. De igual manera se evaluó el comportamiento de la prueba ante interferentes, tales como sangre, orina masculina y femenina, leche materna, secreciones vaginales, materia fecal y saliva. Mostrando resultados positivos para sangre y orina de un individuo con cáncer de próstata se reporta una sensibilidad de 91.6% y una especificidad de 100%.
